BACKGROUND: Thromboembolic event (TEE) rates in the general population and the cancer population are 0.1% to 2% and 10% to 15%, respectively. Our clinical observation is that mesothelioma patients are very susceptible to TEE, including arterial thromboses, but the TEE incidence has not been reported. This retrospective study attempts to determine the rates of TEE in patients with mesothelioma. METHODS: Three hundred seventy-four patients with mesothelioma were identified through the New Mexico SEER database. Sixty-five of them were included in the University of New Mexico tumor registry from 1973 to 2003. Documented TEE rates were abstracted from the patient charts. Chi-square test, Fisher's exact test, and logistic regression were used to identify potentially associated prognostic factors. RESULTS: Fifty-four medical records were reviewed. Patients had a median of six visits (range, 1 to 63 visits). Median age was 60 years (range, 29 to 79 years). Sex distribution was 11 women and 43 men. Anatomic locations of the primary tumor were 35 pleural, 17 peritoneal, 1 pericardial, and 1 pericardial or pleural mesothelioma. The TEE rate was 27.7% (15 of 54), including 10 deep venous thromboses, 2 arterial clots, 2 myocardial infarctions, and 1 pulmonary embolus. No association between the development of TEE and any known prognostic factors were observed. CONCLUSIONS: The 27.7% TEE incidence rate in mesothelioma patients is higher than in other cancer patients. The true incidence of TEE in mesothelioma is likely to be higher than the rate observed in our review, owing to the retrospective nature of the data. Prophylactic anticoagulation trials are recommended to determine the prevention benefit in this high-risk population.

Synthetic triterpenoids, such as 2-cyano-3,12-dioxooleana-1,9-dien-28-oic acid (CDDO) and its derivatives, are an extremely potent class of new anti-cancer therapeutic agents, characterized by high antitumor potency and low toxicity to normal tissues. This report is the first to investigate the effects of C-28 derivatives of CDDO on 22 pediatric solid tumor cell lines, including neuroblastoma, rhabdomyosarcoma, osteosarcoma, and Ewing's sarcoma. We determined IC50s in the range of 5 - 170 nM for inhibition of colony formation and DNA synthesis, and 110 - 630 nM for metabolic cell death and decrease in cell number, using the C-28 CDDO analogs, CDDO methyl ester (CDDO-Me), CDDO imidazolide (CDDOIm), CDDO ethyl amide (CDDO-EA), CDDO trifluoroethyl amide (CDDO-TFEA), and CDDO diethylamide (CDDO-DE). After treatment of human neuroblastoma cells with CDDO-Me, cell cycle studies show depletion of the S-phase, while apoptosis studies show conformational activation and mitochondrial translocation of Bax protein, as well as activation of caspases -3 and -8. These data demonstrate the potential utility of CDDO analogs as promising novel therapeutic agents for high-risk pediatric solid tumors.

Malignant pleural mesothelioma (MPM) is a rare tumour due to occupational asbestos exposure. The incidence of MPM will continue to increase until 2020-2030. The incidence reaches 100 cases/million/year in occupationally exposed populations as opposed to 1 case/million/year in the general population, leading to 800 to 1,000 cases per year in France. The molecular carcinogenesis of MPM is incompletely understood but alterations to genes NF2, c-met, WT1 RASSF and p16 have been described. These genes are involved in cell invasion and motility, cell division and apoptosis control. Histological diagnosis remains difficult and depends on immunohistochemical analysis as described by the French Mesopath group. Clinical diagnosis relies on thoracoscopy and large pleural biopsies, with increasing use of CT-PET for the evaluation of disease extent. Therapeutic strategy includes prophylactic irradiation following drainage or thoracoscopy to prevent tumour nodule development along drainage channels and puncture sites. In selected patients, extensive extra-pleural pneumonectomy can be performed with curative intent. First line chemotherapy is based on a combination of pemetrexed and cisplatin that has demonstrated an improvement in overall survival and quality of life in phase 3 trials. Antiangiogenic agents such as bevacizumab (Avastatin) may be of interest but need to be tested in phase 3 trials. The Mesothelioma Avastatin Pemetrexed Study (MAPS) is ongoing, coordinated by the French Thoracic Cancer Intergroup (IFCT).

Realistic improvement has been recently done for the treatment of malignant pleural mesothelioma. Besides new findings for the epidemiology of the disease, medico-social impact for patients, the knowledge of biological parameters for diagnosis, prognosis and future therapeutic targets as well, the early diagnosis of the disease mainly based on more extended practice of thoracoscopy allows in association with new imaging techniques a careful staging of the disease and consequently new therapeutic implications. Indeed if new balistic assessment of the disease improves the efficacy of radiotherapy and new combined chemotherapy have shown antitumoral responses, surgical strategy takes part in the armamenterium for this disease and combined with others therapeutic modalities seems to be a raisonnable approach despite the lack of prospective, comparative, randomized study and the drawback of current staging. However, the most important point is the multidisciplinary concertation induced by the management of this disease which represents a "model" in thoracic oncology.

 The 2 surgical procedures used are pleurectomy with decortication and extrapleural pneumonectomy.

Surgical resection has been relied upon because radiation and chemotherapy get hold of been ineffective unequivocal treatments. There are now a number of cancer treatment options open to mesothelioma patients. Extrapleural pneumonectomy for selected victims with very early stage disease may expand recurrence-free survival, but the impact it has on overall survival is unknown at this time.

Mesothelioma diagnosis can be intimidating and can scare a lot of people, but mesothelioma diagnosis may give you a fighting chance if can be diagnose early. So do yourself a favor if you think that what you are suffering from and had worked in an asbestos related workplace.

The purpose of such investigations in mesothelioma diagnosis is to confirm and to determine the type of mesothelioma, to 'stage' the disease (measure how severe it is), and so to assess whether the disease is operable.

BACKGROUND: To evaluate pathological complete response rate and to identify the predictor of response after primary systemic chemotherapy (PST) with weekly docetaxel and epirubicin for locally advanced breast cancer. METHODS: Sixty-three patients with locally advanced breast cancer received three cycles PST on day 1 and 8 of each 3-week cycle with epirubicin and docetaxel (epirubicin 45 mg/m(2) intravenous bolus, docetaxel 35 mg/m(2) in 100 ml normal saline infused 1 h), followed by surgery and adjuvant chemotherapy with cyclophosphamide, epirubicin and 5-fluorouracil. The pathological complete response was defined as no invasive carcinoma in breast and axillary nodes after PST. RESULTS: The median tumor sizes (by ultrasound) before and after PST were 6.2 and 2.5 cm, respectively. The negative estrogen receptor (ER) by immunochemical stain was found in 33 (52.4%) patients and HER-2/neu-overexperssion in 12 (19.0%) patients. Clinical overall response rate (ORR) was 89% (95% confidence intervals (95% CI: 81-97), including 38% complete response (95% CI: 26-50), sonographical ORR was 97% (95% CI: 93-100). The pathological complete response were found in 11 patients (18%, 95% CI: 9-27), and 15(24%, 95% CI: 13-35) patients achieved breast only pathological complete response. Nine (27.3%) of thirty-three ER (-) patients and 5 (41.7%) of 12 HER2-positive patients achieved pathological complete response. CONCLUSION: PST with weekly docetaxel and epirubicin were well-tolerated and very high pathological complete response rate was achieved in HER-2/neu -overexpression tumors.

Cholesterol granuloma of the breast is a rare benign condition that can be mistaken for breast cancer. We present a case of a 42-year-old woman who presented with a 1-year history of asymptomatic palpable nodule in the upper external quadrant of the right breast. Mammography and ultrasonography suggested carcinoma, but excisional biopsy revealed cholesterol granuloma with unusual osseous metaplasia. Although reported to occur more frequently in the middle ear and mastoid process, we believe that better awareness of this rare benign condition is most important to avoid misdiagnosis and unnecessary surgery.

Background Meningiomas account for up to 37% of all primary brain tumors. Genetic susceptibility to meningioma is well established, with the risk among relatives of meningioma patients being approximately threefold higher than that in the general population. A relationship between risk of meningioma and exposure to ionizing radiation is also well known and led us to examine whether variants in DNA repair genes contribute to disease susceptibility. Methods We analyzed 1127 tagging single-nucleotide polymorphisms (SNPs) that were selected to capture most of the common variation in 136 DNA repair genes in five case-control series (631 case patients and 637 control subjects) from four countries in Europe. We also analyzed 388 putative functional SNPs in these genes for their association with meningioma. All statistical tests were two-sided. Results The SNP rs4968451, which maps to intron 4 of the gene that encodes breast cancer susceptibility gene 1-interacting protein 1, was consistently associated with an increased risk of developing meningioma. Across the five studies, the association was highly statistically significant (trend odds ratio = 1.57, 95% confidence interval = 1.28 to 1.93; P(trend) = 8.95 x 10(-6); P = .009 after adjusting for multiple testing). Conclusions We have identified a novel association between rs4968451 and meningioma risk. Because approximately 28% of the European population are carriers of at-risk genotypes for rs4968451, the variant is likely to make a substantial contribution to the development of meningioma.

The ordered, directional migration of T-lymphocytes is a key process during immune surveillance, immune response, and development. A novel series of pyrrolo-1,5-benzoxazepines have been shown to potently induce apoptosis in variety of human chemotherapy resistant cancer cell lines, indicating their potential in the treatment of both solid tumors and tumors derived from the hemopoietic system. Pyrrolobenzoxazepine 4-acetoxy-5-(1-naphtyl)naphtho[2,3-b]pyrrolo[1,2-d][1,4]-oxazepine (PBOX-15) has been shown to depolymerize tubulin in vitro and in the MCF7 breast cancer cell line. We hypothesized that this may suggest a role for this compound in modulating integrin-induced T-cell migration, which is largely dependent on the microtubule dynamics. Experiments were performed using human T lymphoma cell line Hut78 and peripheral blood T-lymphocytes isolated from healthy donors. We observed that human T-lymphocytes exposed to PBOX-15 have severely impaired ability to polarize and migrate in response to the triggering stimulus generated via cross-linking of integrin lymphocyte function associated antigen-1 receptor. Here, we show that PBOX-15 can dramatically impair microtubule network via destabilization of tubulin resulting in complete loss of the motile phenotype of T-cells. We demonstrate that PBOX-15 inhibitory mechanisms involve decreased tubulin polymerization and its post-translational modifications. Novel microtubule-targeting effects of PBOX-15 can possibly open new horizons in the treatment of overactive inflammatory conditions as well as cancer and cancer metastatic spreading.

BACKGROUND: Laparoscopic colorectal resection has become popular. The recently developed da Vinci Surgical System promises to facilitate endoscopic surgery and overcome its disadvantages. This study therefore aimed to compare the short-term results between robotic tumor-specific mesorectal excision (R-TSME) using the da Vinci Surgical System and conventional laparoscopic tumor-specific mesorectal excision (L-TSME) in rectal cancer patients. METHODS: Between April 2006 and February 2007, 36 patients were randomly assigned to receive R-TSME or L-TSME. During the study, 18 patients underwent robotic low anterior resection using the da Vinci Surgical System, and 18 patients had conventional laparoscopic low anterior resection. Patient characteristics, perioperative clinical results, complications, and pathologic details were compared between the two groups. RESULTS: The patient characteristics were not significantly different between the two groups. The mean operating time, hemoglobin change, and conversion rate were not significantly different between the groups. Complications were treated conservatively and did not require surgical intervention in the R-TSME group. BACKGROUND AND OBJECTIVES: Approximately 10-40% of colorectal cancer patients with potentially resectable hepatic metastases are incorrectly deemed resectable on standard pre-operative evaluation, including contrast-enhanced CT. Laparoscopy can identify unresectability in a majority of patients at highest risk of being incorrectly deemed resectable, sparing them an unnecessary laparotomy. However, laparoscopy requires an added investment by surgeons, patients, and payers. This analysis seeks to ascertain whether that investment is cost-effective. METHODS: A decision tree model was developed to evaluate the societal cost-effectiveness of laparoscopy versus laparotomy in colorectal cancer patients with hepatic metastases deemed resectable on standard pre-operative evaluation. This comparison involved the cost, the effectiveness, and the incremental cost-effectiveness (the cost in dollars for each quality-adjusted life-year saved) of each option. Sensitivity analysis was performed to evaluate the model's validity under a variety of assumptions. RESULTS: The cost-effectiveness of performing laparoscopy prior to laparotomy for resection of colorectal hepatic metastases depends primarily upon the probability of resectability determined at laparoscopy, and on the sensitivity of diagnostic laparoscopy. CONCLUSION: Laparoscopy for initial evaluation of resectability of hepatic metastases from colorectal cancer is most likely to benefit patients and save costs when performed after pre-operative risk stratification in patients at high risk of radiographically occult unresectable disease. Oncogenic K-ras mutations are frequently observed in colon cancers and contribute to transformed growth. Oncogenic K-ras is detected in aberrant crypt foci (ACF), precancerous colonic lesions, demonstrating that acquisition of a K-ras mutation is an early event in colon carcinogenesis. Here, we investigate the role of oncogenic K-ras in neoplastic initiation and progression. Transgenic mice in which an oncogenic K-ras(G12D) allele is activated in the colonic epithelium by sporadic recombination (K-rasLA2 mice) develop spontaneous ACF that are morphologically indistinguishable from those induced by the colon carcinogen azoxymethane (AOM). Similar neoplastic changes involving the entire colon are induced in transgenic mice constitutively expressing K-ras(G12D) throughout the colon (LSL-K-ras(G12D)/Villin-Cre mice). However, the biochemistry and fate of K-ras-induced lesions differ depending upon their location within the colon in these mice. In the proximal colon, K-ras(G12D) induces increased expression of procarcinogenic protein kinase CbetaII (PKCbetaII), activation of the MEK/ERK signaling axis and increased epithelial cell proliferation. In contrast, in the distal colon, K-ras(G12D) inhibits expression of procarcinogenic PKCbetaII and induces apoptosis. Treatment of K-rasLA2 mice with AOM leads to neoplastic progression of small ACF to large, dysplastic microadenomas in the proximal, but not the distal colon. Thus, oncogenic K-ras functions differently in the proximal and distal colon of mice, inducing ACF capable of neoplastic progression in the proximal colon, and ACF with little or no potential for progression in the distal colon. Epigenetics is concerned about heritable changes in gene expression without alteration of the coding sequence. Epigenetic modification of chromatin includes methylation of genomic DNA as well as post-translational modification of chromatin-associated proteins, in particular, histones. The spectrum of histone and non-histone modifications ranges from the addition of relatively small groups such as methyl, acetyl and phosphoryl groups to the attachment of larger moieties such as poly(ADP-ribose) and small proteins ubiquitin or small ubiquitin-like modifier (SUMO). The combinatorial nature of DNA methylation and histone modifications constitutes a significant pathway of epigenetic regulation and considerably extends the information potential of the genetic code. Chromatin modification has emerged as a new fundamental mechanism for gene transcriptional activity control associated with many cellular processes like proliferation, growth, and differentiation. Also it is increasingly recognized that epigenetic modifications constitute important regulatory mechanisms for the pathogenesis of malignant transformations. We review here the recent progress in the development of chemical inhibitors/activators that target different chromatin modifying enzymes. Such potent natural or synthetic modulators can be utilized to establish the quantitative contributions of epigenetic modifications in DNA regulated pathways including transcription, replication, recombination and repair, as well as provide leads for developing new cancer therapeutics. RESULTS: Methylation frequencies in the tumor samples were detected in 18% of samples for the breast cancer 1 gene BRCA1, in 43% of samples for the tissue inhibitor of metalloproteinase 3 gene TIMP-3, in 38% of samples for the cyclin-dependent kinase inhibitor 4A gene p16INK4a, in 54% of samples for the cadherin 13 gene CDH13, in 50% of samples for the death-associated protein kinase gene DAPK, in 11% of samples for the E-cadherin gene ECAD, in 25% of samples for the insulin-like growth factor binding protein 7 gene IGFBP7, in 18% of samples for the Ras association domain family 1 gene RASSF1, in 68% of samples for the adenomatous polyposis coli gene APC, in 7% of samples for the cyclin-dependent kinase inhibitor gene p15, in 18% of samples for the CD44 cell adhesion molecule gene, in 29% of samples for the human Mut-L homolog gene hMLH, in 32% of samples for the human telomerase reverse transcriptase gene hTERT, in 64% of samples for the calcitonin gene-related polypeptide alpha gene CALCA, and in 54% of samples for the estrogen receptor gene ER; however, methylation was not observed in the majority of corresponding nonmalignant tissues. Six samples in from 28 tumors had >6 genes methylated, and 1 sample had 13 genes methylated. Methylation of these genes was correlated with some clinicopathologic patient characteristics. CONCLUSIONS: This study demonstrated that a 3-D microarray could be used to detect DNA hypermethylation and provided a high-throughput platform for DNA hypermethylation analysis. Cancer 2008. Mesothelioma Cancer   

Unlike lung cancer, there is no association between mesothelioma and smoking Mesothelioma is a wizardry of cancer that is almost always caused by previous exposure to asbestos Mesothelioma is the word used to describe a cancerous tumor that involves the mesothelial cells of an organ, often the lungs, heart, or abdominal organs. Its greatest easily understood site is the pleura (outer lining of the lungs and chest cavity), but it may also occur in the peritoneum (the lining of the abdominal cavity) or the pericardium (a sac that surrounds the heart).

 Surpassingly people who develop mesothelioma get hold of worked on jobs where they inhaled asbestos particles, or carry been exposed to asbestos dust and fibre in other ways, such as by washing the clothes of a family member who worked with asbestos, or by home renovation using asbestos cement products. It can also occur in children; however, these cases are not thought to be associated with asbestos exposure.

 Symptoms of peritoneal mesothelioma include weight loss and cachexia, abdominal swelling and pain due to ascites (a buildup of fluid in the abdominal cavity).  A history of exposure to asbestos may increase clinical suspicion for mesothelioma. Other symptoms of peritoneal mesothelioma may include bowel obstruction, blood clotting abnormalities, anemia, and fever. Symptoms of mesothelioma may not appear until 20 to 50 years after exposure to asbestos. This is the end of a expertise that usually begins with symptoms that send most people to the doctor: a fluid build-up around the lungs (pleural effusions), shortness of breath, suffering in the chest, or suffering or swelling in the abdomen.

 A physical examination is performed, followed by chest X-ray and often lung function tests. Shortness of breath, cough, and aches and in the chest due to an accumulation of fluid in the pleural space are often symptoms of pleural mesothelioma.

 If the cancer has body beyond the mesothelium to other parts of the spread, symptoms may include aches and, trouble swallowing, or swelling of the neck or make contact with.

Tissue is removed, placed under the microscope, and a pathologist makes a definitive diagnosis, and issues a pathology report. Malignant mesothelioma is a rare type of cancer in which malignant cells are found in the sac lining the chest or abdomen. Extremely malignant mesotheliomas accept complex karyotypes, with extensive aneuploidy and rearrangement of swarms chromosomes. 

 

Most people with malignant mesothelioma require worked on jobs where they breathed asbestos. Exposure to airborne asbestos particles increases one's risk of developing malignant mesothelioma.

 Malignant mesothelioma has a peak incidence 35-45 years after asbestos exposure. Malignant mesothelioma has also been linked to remedial radiation using thorium dioxide and zeolite, a silicate in the soil.

 Malignant mesothelioma is often just called simply Mesothelioma and is a form of lung cancer that is quite rare.

 

Malignant mesothelioma is more explicit in men, with a male-to-female ratio of 3:1. In this disease, malignant cells develop in the mesothelium, a protective lining that covers extremely of the body's internal organs.

 

The be obliged of radiation therapy in pleural mesothelioma has been shown to alleviate aches and in the majority of patients that are treated. But unfortunately, the duration of symptom control is short-lived. Radiation has no effect on survival, but it has caused significant palliation in 50% of patients treated for chest aches and and chest wall metastasis. There are now a number of cancer treatment options open to mesothelioma victims. Extrapleural pneumonectomy for selected victims with very early stage disease may extend recurrence-free survival, but the impact it has on overall survival is unknownat this time.

 

Surgical resection has been relied upon because radiation and chemotherapy set up been ineffective legitimate treatments. The 2 surgical procedures used are pleurectomy with decortication and extrapleural pneumonectomy.

The patient was a 66-year-old man. Total gastrectomy was performed due to BorrmannVtype moderately differentiated adenocarcinoma of the middle part of the stomach. The final diagnosis was UM 7x6 cm, sT3, sN1, sH0, sP0, sM0, sCY0, sStage IIIA, PM (-), DM (-), D2+alpha, Cur B, ly2, v2. Because CEA had increased slowly from ten months after the operation, it was judged a possibly partial relapse. Twice-administered CDDP 80 mg/body and S-1 80 mg on the eighth day served to decrease CEA. He was hospitalized again due to fracture of the spine, though he left the hospital once. The patient was diagnosed by MRI inspection of the vertebrae thoracicae and the lumbar vertebra as multiple spinal bone metastases. After CDDP 60 mg/body of day 8 was administered twice at S-1 80 mg/day, CEA became normal. Osteolytic changes of the spinal bone disappeared. The 24 months from February 2004 to January 2006 passed without additional treatment. His CEA value tended to rise within the normal range in February 2006. After that, the CEA level increased with a 4-week cycle of 2 weeks of S-1 40-80 mg alternating with a two-week rest.

Trastuzumab/capecitabine combination therapy was performed for two advanced/recurrent breast cancer cases with acute deterioration of the disease. The time until partial response in the first case was 7 months and in the second case 12 months. Adverse events were slight, though a standard dose as a single drug was used for each drug. The treatment time course was shortened because capecitabine was an oral drug. Therefore, this combination therapy was expected to maintain high quality of life and obtain a high response rate.

A 77-year-old woman who underwent a right modified radical mastectomy 23 years ago with no further adjuvant treatment presented with a right chest wall mass (3x4x2 cm) at the scar. She had no symptoms nor metastasis. The laboratory data were normal including tumor marker. The mass was diagnosed as compatible with a local recurrence tumor from the previous breast cancer on the ultrasonography and chest CT. After obtaining her informed consent for the therapy, we performed 60 Gy/30 fr radiotherapy on the recurrent tumor on her right chest wall with concurrent oral chemo-endocrine therapy. The first regimens were tamoxifen 20 mg/day and 5'-DFUR 600 mg/day, followed by tore- mifene 80 mg/day and 5'-DFUR 600 mg/day, and then the tumor disappeared. But three years later, we found tumor regrowth. We changed the regimen, giving 5'-DFUR 1,200 mg/day and cyclophosphamide 100 mg/day for 2 weeks followed by a 1-week drug-free period, then added 10 Gy/5 fr radiotherapy and hyperthermia twice a week. Final regimens were anastrozole 1 mg/day and capecitabine 900 mg/day. The recurrent tumor decreased and the disease stabilized. After these therapies, she had very good quality of life. We recommend radiation and/or hyperthermia with concurrent oral chemo-endocrine therapy as useful for the delayed recurrence of elderly breast cancer after a modified radical mastectomy.

PURPOSE:: To examine the feasibility of offering genetic susceptibility testing for lung cancer (GSTM1) via the Internet to smokers who were blood relatives of patients with lung cancer. Outcomes include proportion who logged on to the study website to consider testing, made informed decisions to log on and to be tested. METHODS:: Baseline measures were assessed via telephone survey. Participants could choose to log on to the study website; those who did were offered testing. Informed decisions to log on and to be tested were indicated by concordance between the decision outcome and test-related attitudes and knowledge. RESULTS:: Three hundred four relatives completed baseline interviews. One hundred sixteen eligible relatives expressed further interest in receiving information via the web. Fifty-eight logged on and 44 tested. Those logging on expressed greater quit motivation, awareness of cancer genetic testing, and were more likely to be daily Internet users than those who did not log on. Approximately half of the sample made informed decisions to log on and to be tested. CONCLUSION:: Interest in a web-based protocol for genetic susceptibility testing was high. Internet-delivered decision support was as likely as other modalities to yield informed decisions. Some subgroups may need additional support to improve their decision outcomes.

PURPOSE OF REVIEW: To examine recent data about the association between rheumatic disorders and cancer. This article focuses on paraneoplastic rheumatic disorders, which usually precede by a short period of time the diagnosis of malignancy, and on malignant transformation, which occurs late in the course of rheumatic disorders. Evidence of causality between malignancies and rheumatic disorders was reviewed based on statistical indicators (standardized incidence ratios and odds ratios) and by applying Bradford Hill's criteria of causality. RECENT FINDINGS: Firm epidemiological evidence was found attesting that dermatomyositis and polymyostis may present as paraneoplastic syndromes. Several other musculoskeletal disorders may be present akin to paraneoplastic syndrome, based on clinicians' impressions, but with scarce epidemiological evidence supporting a causal determinism. In contrast, robust evidence has accumulated on the role of longstanding rheumatoid arthritis, Sjögren's syndrome and systemic sclerosis as premalignant conditions. Evidence that systemic lupus erythematosus may evolve into lymphoma is equivocal. SUMMARY: The link between malignancies and rheumatic disorders may impact on clinical practice. First, paraneoplastic rheumatic syndromes can provide the clinician with hints for earlier diagnosis of occult cancer. Second, the risk of malignant transformation during the course of rheumatic disorders may motivate the search for strategies aimed at prevention.
